Recommended Procedure

  • Label the top of the vials
  • Take an aseptic milk sample direct from the cow after cleaning the teat end with alcohol
  • If pipetting from a previously collected sample, use a sterile pipette to instil 3 drops into well in centre of tube.
  • Flick downwards once to ensure milk is dispersed deep into the well.
  • Tap excess milk out of the top before closing the lid firmly
  • Label the Overnighter with the cow number and the quarter sampled
  • Incubate at a temperature that is close to body temperature for best results such as on the water heater, on chiller heat exchange unit or in an inside pocket. “Top down” on the hot water cupboard is best
  • Start watching for colour change after 8-12 hours. Always read in good light If there is no change, reincubate for a further 12-24 hours. (see

  • Where bacterial numbers are low, incubation may take up to 48 hours.
